From Interstate 680 in Alameda County, exit Calaveras (exit 21a). Follow the brown "parks" signs to Sunol Regional Wilderness.

Description and other notes:
Follow the Ohlone Wilderness Trail from Sunol HQ (junction #13 on the map). In Sunol Wilderness area, the trail's segment is called McCorkle Trail for 2+ miles. Soon after Sunol Backpack Area, Ohlone Wilderness Trail leads into Ohlone Regional Wilderness. Follow it to point #28 on the map. Go straight from #28 to Rose Peak, which is about 0.2 miles off the Ohlone Trail, and at 3817', is the highest publicly accessible peak in Alameda County. From there, you can take the same Ohlone Trail (marked in red on the map and at trail junctions) back to the parking lot. The hike is on the strenuous side (but from Livermore, it is even more strenuous) but worth it because it provides great exercise and great views.
